The pharmacology of halogenated salicylanilides and their anthelmintic use in animals

The halogenated salicylanilides are a large group of compounds developed mainly for their antiparasitic activity in animals. Several halogenated salicylanilides with potent antiparasitic activity have been synthesised of which only closantel, niclosamide, oxyclozanide, rafoxanide and Resorantel are commercially available. Closantel and rafoxanide, which represent the most important drugs in the group, are used extensively for the control of Haemonchus spp. and Fasciola spp. infestations in sheep and cattle and Oestrus ovis in sheep in many parts of the world. Niclosamide is used extensively for its anticestodal activity in a wide range of animals. Antiparasitic activity of the halogenated salicylanilides has also been demonstrated against a large number of other internal parasites, in particular haematophagous helminths, and external parasites including ticks and mites, in a variety of animal species. Several cases of toxicity and mortality have been reported for closantel and rafoxanide in sheep and goats. Their unique pharmacokinetic behaviour appears to play an important role in the efficacy and safety of these compounds. The chemical and physical characteristics, mode of action, pharmacokinetics, antiparasitic activity and toxicity of the halogenated salicylanilides in animals are reviewed.

The anthelmintic efficacy of Resorantel against Houttuynia struthionis in ostriches

Resorantel at a dosage rate of 130 mg kg-1 live body mass was found to be highly effective against Houttuynia struthionis in ostriches when dosed alone or in combination with fenbendazole or levamisole.

The metabolism of the anthelmintics clioxanide and Resorantel and related compounds in vitro by Moniezia expansa, Ascaris suum and mouse- and sheep-liver enzymes

1. Clioxanide and related compounds were deacetylated by the cestode Moniezia expansa, the nematode Ascaris suum, by enzymes prepared from these species and by mouse- and sheep-liver homogenates. Deacetylase activity was found in the cytosol of cestode proglottids throughout the strobila, and in the cytosol of nematode intestinal cells and reproductive tract. 2. The O-deacetylases from both helminths showed similar pH optima of about 7.0. Activity was enhanced by Ca2+ and low molecular weight thiols. Cu2+, Cd2+, Hg2+, Zn2+, and La2+ inhibited the deacetylation of clioxanide. 3. Resorantel and clioxanide were not hydrolysed at the amide bond by helminth or mammalian enzymes. 4. Resorantel was hydroxylated by mammalian microsomal enzymes, but helminths did not modify the molecule.

The anthelmintic efficacy of Resorantel against cestodes in sheep

The anthelmintic efficacy of Resorantel against Thysaniezia giardi and Avitellina spp. was tested in sheep at a dosage rate of 65 mg kg-1 live mass. The reduction obtained was 100% in at least 80% of the treated flock.

Chemotherapy of paramphistomosis in sheep

Controlled trials were used to assess the efficacy of various anthelmintics against immature and adult paramphistomes in 75 experimentally or naturally infected sheep. Albendazole (20 mg/kg), praziquantel (10 mg/kg), nitroxynil (10 mg/kg) triclabendazole (10 and 100 mg/kg), profenophos (25 mg/kg) and netobimin (15 mg/kg) had little or no activity against adult or immature fluke. Niclosamide at 100 mg/kg had high efficacy (99%) against intestinal fluke but none against adult fluke. A 2- tertiary-butyl benzthiazole compound (CGA 72630) at 25 mg/kg and Resorantel at 65 mg/kg had very high efficacy against both adult and immature fluke in the rumen and small intestine respectively. The efficacy of other anthelmintics which have been used against paramphistomes in sheep is reviewed.
