Isodeoxycholic Acid is a secondary bile acid [1]. Isodeoxycholic Acid regulates bile acid synthesis, enterohepatic circulation, and energy metabolism, influencing the composition of the intestinal microbiome and inflammatory responses [2]. Isodeoxycholic Acid is primarily used to treat cholestasis, hepatobiliary diseases, and metabolic syndrome [3-4].
In CD4⁺T cells, Isodeoxycholic Acid (100μM; 3d) acts on dendritic cells, weakening their immunostimulatory properties and thereby enhancing the induction of Foxp3 [5].

Preparation Method | Naïve CD4⁺T cells (CD44⁻CD62L⁺GFP⁻) were isolated from the spleen and peripheral lymph nodes of Foxp3-GFP mice and co-cultured with dendritic cells (1×105). They were stimulated with Isodeoxycholic Acid (diluted from a 100mM DMSO stock solution) and anti-CD3ε antibody (1μg/mL). T cell proliferation (CellTrace Violet labeling), Foxp3 induction (GFP reporter gene), and cytokine production (PMA/Ionomycin heavy stimulation, Brefeldin A/Monensin secretion blockade) were assessed under different polarization conditions (Treg induction with TGF-β/IL-2 and Th17 induction with TGF-β/IL-6). Flow cytometry was used to quantify cell number using internal control beads to investigate the regulatory effects of Isodeoxycholic Acid on CD4⁺T cell differentiation and function. |
