Description

TOTO 1 is an excellent nuclear and chromosomal counterstain that cannot penetrate live cells, but can penetrate dead cells with damaged membrane features, making it a useful dead cell indicator within cell populations. TOTO 1 has almost no fluorescence in an environment without nucleic acid. Once it binds to nucleic acid, it produces bright green fluorescence with maximum excitation and emission wavelengths of~514/531nm, respectively

Important reminder (before purchase or initial use): Please make sure to refer to:
1、 Precautions for preparing storage solutions of fluorescent dyes (in powder form, especially oxygen sensitive probes)
1) Fluorescent probes are very stable in solid (powder) state and can be stored at the required temperature according to the instructions. They can be used within the expiration date.
2) Fluorescent probes are dissolved in organic solvents such as DMSO and prepared into storage solutions. Generally, they are stored at -20 ° C (for use within 2-3 months, or even shorter, please consult for details); Store at -80 ° C (for use within 6 months, please consult). The premise is that the organic solvents used must be of high quality and anhydrous, especially DMSO, which must be freshly opened.
3) The prepared storage solution must be stored in a low capacity cryovial with good sealing and a screw cap (EP tubing is not allowed), and at least 5-10 U I/tube should be divided and stored away from light.
4) For certain special compounds (sensitive to air or with unstable structures), the storage period may be particularly short, and they can only be used on the same day. These compounds will be explained in the instruction manual.

2、 Precautions for fluorescent dyes (provided in the form of storage solutions dissolved in organic solvents)
1) Fluorescent probes in the form of storage solutions dissolved in organic solvents are relatively stable compounds; But when receiving such products, users also need to pack them according to the single dosage (5-10ul/tube) and store them in a sealed and dark place below -20 ℃ to reduce the number of repeated freeze-thaw cycles.
2) Please make sure to store in a low capacity cryovial with good sealing and screw cap (EP tubing is not acceptable), and avoid light exposure.

重要提醒(购买或初次使用前)请务必查阅:
一、荧光染料(粉末形式,特别是对氧敏感探针)配制储存液的注意事项
1)荧光探针在固体(粉末)状态很稳定,按照说明书要求温度来保存,效期内使用即可。
2)荧光探针用有机溶剂比如:DMSO,溶解配制成储存液之后,一般来说,放到-20℃保存(2-3个月内使用,甚至更短,具体可咨询);放到-80°C保存(6个月内使用,具体可咨询)。前提是,使用的有机溶剂必须是高质量且无水的,特别是DMSO,必须是新鲜开封。
3)配制的储存液请务必用密封性好且螺旋盖的低容量冻存管保存(不可用EP管),至少按照5-10u I/管来分装,避光冻存。
4)对于某些特殊化合物(对空气敏感或存在不稳定结构),可能保存周期特别短,甚至只能当天使用。这些化合物说明书上会有说明。

二、荧光染料 (以溶于有机溶剂的储存液形式提供)的注意事项
1) 以溶于有机溶剂的储存液形式的荧光探针相对来说是比较稳定的化合物;但收到这类产品,也需用户根据单次用量(5-10ul/管来分装),-20℃以下密封避光保存,减少反复冻融次数。
2) 请务必用密封性好且螺旋盖的低容量冻存管保存(不可用EP管)。务必避光。

实验参考方法

染色步骤
[注意]:以下步骤适用于大多数细胞类型。生长培养基、细胞密度、是否存在其它细胞类型都可能影响染色。玻璃表面残留的去污剂也可能影响许多有机体的染色结果,在含或不含细胞的溶液内都可能看到明亮的染色材料。
1)按照实验需求培养和处理细胞。
2)吸走培养基。
3)加入TOTO 1工作液 (1-10μM)到细胞内(或悬浮细胞,或贴壁细胞),染色15-60min。
[注①]于实验前将低温保存的储存液TOTO 1 (5mM in DMSO)置于室温(25℃)或温育片刻使其充分融化,低速离心后,吸取适量储存液用HHBS缓冲液或其它生理缓冲液稀释到工作浓度。第一次使用储存液的时候可根据单次用量分装,≤-20C避光干燥保存,降低反复冻融次数。
[注②]第一次实验务必尝试几种染色浓度,以确定最佳的工作浓度。高染料浓度很可能引起其它细胞结构的非特异染色。
4)吸走染色工作液,加入HHBS缓冲液或其它适宜的缓冲
