Antimicrobial evaluation of imines and thiazolidinones derived from 3-phenylpropane hydrazide

Methyl 3-phenylpropanoate (1), after hydrazination into 3-phenylpropanehydrazide (2), was converted into N-arylidene-3-phenylpropane hydrazides (3a-e), which on cyclization with thioglycolic acid yielded N-(4-oxo-2-arylthiazolidin-3-yl)-3-phenylpropanamides (4a-e). All the proposed structures of newly synthesized compounds were in full agreement with the spectral data. Due to para substitution, compound 3a, 4a and 4b were found to be the most potent when evaluated for antibacterial and antifungal activities.
Structure-activity relationships of phenylpropanoids as antifeedants for the pine weevil Hylobius abietis

Ethyl cinnamate has been isolated from the bark of Pinus contorta in the search for antifeedants for the pine weevil, Hylobius abietis. Based on this lead compound, a number of structurally related compounds were synthesized and tested. The usability of the Topliss scheme, a flow diagram previously used in numerous structure-activity relationship (SAR) studies, was evaluated in an attempt to find the most potent antifeedants. The scheme was initially followed stepwise; subsequently, all compounds found in the scheme were compared. In total, 51 phenylpropanoids were tested and analyzed for SARs by using arguments from the field of medicinal chemistry (rational drug design). Individual Hansch parameters based on hydrophobicity, steric, and electronic properties were examined. The effects of position and numbers of substituents on the aromatic ring, the effects of conjugation in the molecules, and the effects of the properties of the parent alcohol part of the esters were also evaluated. It proved difficult to find strong SARs derived from single physicochemical descriptors, but our study led to numerous new, potent, phenylpropanoid antifeedants for the pine weevil. Among the most potent were methyl 3-phenylpropanoates monosubstituted with chloro, fluoro, or methyl groups and the 3,4-dichlorinated Methyl 3-phenylpropanoate.
Methoxycarbonylation of olefins catalyzed by palladium complexes bearing P,N-donor ligands

The methoxycarbonylation of alkenes catalyzed by palladium(II) complexes with P,N-donor ligands, 2-(diphenylphosphinoamino)pyridine (Ph2PNHpy), 2-[(diphenylphosphino)methyl]pyridine (Ph2PCH2py), and 2-(diphenylphosphino)quinoline (Ph2Pqn) has been investigated. The results show that the complex [PdCl(PPh3)(Ph2PNHpy)]Cl or an equimolar mixture of [PdCl2(Ph2PNHpy)] and PPh3, in the presence of p-toluensulfonic acid (TsOH), is an efficient catalyst for this reaction. This catalytic system promotes the conversion of styrene into methyl 2-phenylpropanoate and Methyl 3-phenylpropanoate with nearly complete chemoselectivity, 98% regioselectivity in the branched isomer, and high turnover frequency, even at alkene/Pd molar ratios of 1000. Best results were obtained in toluene-MeOH (3 : 1) solvent. The Pd/Ph2PNHpy catalyst is also efficient in the methoxycarbonylation of cyclohexene and 1-hexene, although with lower rates than with styrene. Related palladium complexes [PdCl(PPh3)L]Cl (L = Ph2PCH2py and Ph2Pqn) show lower activity in the methoxycarbonylation of styrene than that of the 2-(diphenylphosphinoamino)pyridine ligand. Replacement of the last ligand by (diphenylphosphino)phenylamine (Ph2PNHPh) or 2-(diphenylphosphinoaminomethyl)pyridine (Ph2PNMepy) also reduces significantly the activity of the catalyst, indicating that both the presence of the pyridine fragment as well as the NH group, are required to achieve a high performing catalyst. Isotopic labeling experiments using MeOD are consistent with a hydride mechanism for the [PdCl(PPh3)(Ph2PNHpy)]Cl catalyst.
