Description

Iron dextran is an injectable complex comprising ferric hydroxide and a low-molecular-weight fraction of dextran. Iron dextran is a type of intravenous iron agent specifically used for the treatment of iron deficiency anemia[1].

In vitro, Iron dextran(1.9–4000μg/ml; 24h) exhibited certain cytotoxicity to HepG2 cells, with a half - inhibitory concentration (IC50) of 1178μg/ml. When used in combination with TQ (thymoquinone), TQ at a specific dose would enhance its toxicity[2].

In vivo, Iron dextran (200mg/kg/every other day; 1,2,3 or 4 weeks; i.p.) induces iron overload and CTSB/CTSD up-regulation in the liver and spleen of mice[3]. Iron dextran (0, 1.1, 3.3 and 9.9mg/(100g·bw·d); 4 weeks; i.g.) played an important role in improving iron deficiency anemia, promoting vitamin D metabolism and activation, regulating iron metabolism balance, and promoting growth and development in rats[4].

在体外,Iron dextran (1.9-4000μg/ml; 24h)对HepG2细胞表现出一定的细胞毒性,半数抑制浓度(IC50)为1178μg/ml。当与TQ(牛蒡子苷)联合使用时,TQ在特定剂量下会增强其毒性[2]

在体内,Iron dextran (200mg/kg/隔一天; 1、2、3和4周; 腹腔注射)可诱导小鼠肝脏和脾脏中铁过载以及组织蛋白酶B(CTSB)和D(CTSD)的上调[3]。Iron dextran (0、1.1、3.3和9.9mg/(100g·bw·d); 4周; 腹腔注射)在改善大鼠缺铁性贫血、促进维生素D代谢和活化、调节铁代谢平衡以及促进生长发育等方面发挥了重要作用[4]

Preparation Method

The HepG2 cell line was cultured in 25cm² flasks containing HG-DMEM medium supplemented with 1% penicillin-streptomycin and 10% fetal bovine serum. The cells were maintained in a humidified atmosphere of 5% CO₂ at 37°C. For cytotoxicity experiments, HepG2 cells were seeded at a density of 10,000 viable cells per well in sterile 96-well plates and incubated overnight at 37°C and 5% CO₂. In the first step, the cells were treated with different concentrations of TQ (0.5-300×10³nM) and Iron dextran (1.9-4000µg/ml) for 24 hours, and the cell viability was determined using the MTT method separately. In the next step, HepG2 cells were pretreated with safe doses of TQ (1.8-15×10³nM) before being exposed to various concentrations of Iron dextran. After 24 hours, 0.5mg/ml of MTT reagent was added to each well and incubated for 4 hours. Finally, 100µl of dimethyl sulfoxide (DMSO) was added to each well to dissolve the formazan crystals, and the changes in absorbance were measured at 570nm using a microplate reader.

Preparation Method

Eight-week-old mice were housed in a near pathogen-free environment (temperature 20°C ± 2°C, humidity 50%–60%, 12h light/dark cycle), with four animals per polypropylene cage and food and water provided ad libitum. Mice were intraperitoneally injected with 200mg/kg Iron dextran every other day for 1, 2, 3, or 4 weeks. Control mice were injected with the same amount of saline.

Dosage form

200mg/kg; 1, 2, 3, or 4 weeks; i.p.

Applications

Iron dextran induces iron overload and CTSB/CTSD up-regulation in the liver and spleen of mice.
