Cyclo(Arg-Gly-Asp-D-Phe-Cys), commonly abbreviated as cRGDfC, is a cyclic RGD peptide that shows high affinity and selectivity for the integrin αvβ3 [1]. Treatment of murine embryonic stem cells (mESCs) with Cyclo(Arg-Gly-Asp-D-Phe-Cys) peptide can induce some cells to express early ectoderm lineage markers [2]. Cyclo(Arg-Gly-Asp-D-Phe-Cys) is mainly used to modify liposomes and other delivery systems to achieve targeted drug delivery to tumor tissues [3-4].
In murine embryonic stem cells, Cyclo(Arg-Gly-Asp-D-Phe-Cys) (0.5mM; 24 h) down-regulated the transcription factors Oct 4, Sox 2, and Nanog [2].

Preparation Method | Murine embryonic stem cells (mESCs) were embedded in a type I collagen matrix. Briefly, mESCs at passage 8 to 10 were trypsinized and re-suspended in β-glycerol phosphate medium (βGP). βGP medium consisted of high glucose 5 × DMEM powder medium supplemented with 10mM βGP and with FBS, NEAA, penicillin-stereptomycin solution, and βME equal to the standard cell culture procedure. To prepare one collagen construct, 1,000,000 cells were re-suspended in 200μL βGP medium and mixed with 800μL of bovine collagen type I solution (3mg/mL) and of 20μL sodium hydroxide. For the integrin studies, Cyclo(Arg-Gly-Asp-D-Phe-Cys) (cRGDfC) peptide was dissolved in 3% citric acid and this was supplemented to mESC-Col I solution to achieve a final concentration of 0.5mM cRGDfC. Finally, mESC-Col I solution, in the presence and absence of cRGDfC peptide, was pipetted into a 12 well plate and allowed to polymerize in a humidified incubator with 5% CO2 at 37℃. |
