FITC-Dextran (MW 70000) is a fluorescein isothiocyanate (FITC) dextran fluorescent probe with Ex/Em=495nm/525nm[1]. In vitro, FITC-Dextran (MW 70000) can be used to study cell permeability, such as the determination of blood-brain barrier permeability and the degree of blood-brain barrier damage[2, 3]. In vivo, FITC-Dextran (MW 70000) can be used to measure endothelial permeability in ischemia/reperfusion models[4]. FITC-Dextran (MW 70000) can be used as a tracer for the movement of macromolecules in the nervous system[5]. FITC-Dextran (MW 70000) can be used to study the encapsulation and release capacity, selectivity and efficiency of drug delivery vehicles[6].

1. 溶液配制
(1)工作液:用实验缓冲液溶解FITC-Dextran (MW 70000),配制所需浓度的工作液,例如:0.1mg/mL。
注意:最佳的工作浓度请根据实际情况调整或参阅文献自行设置梯度浓度进行摸索。工作液必须现配现用。
2. FITC-Dextran (MW 70000)体外血脑屏障通透性测定[1](来自文献,仅做参考)
体外血脑屏障通透性研究采用transwell法。
(1)人脑微血管内皮细胞(HBMEC)接种于孔径为0.4μm的transwell系统中,放置于24孔板中。
(2)在transwell单层形成后,将transwell和下腔的培养基分别替换为条件培养基和对照培养基,孵育过夜。
(3)上腔加入FITC-Dextran (MW 70000)(0.1mg/mL)。孵育20min后检测荧光穿透。血管完整性评价为transwell和下腔之间的吸光度差异。
注意:该实验方案为使用FITC-Dextran (MW 70000)测定血脑屏障通透性提供指导。可以根据其他文献和具体实验要求进行调整。
