Preparation method |
Osteoblasts at passage 3 were used to perform all cell studies and were divided into six groups, which were cultured in the presence of various concentrations of 5-HT : 0 M (control group) and 1 nM, 10 nM, 100 nM, 1 µM, and 10 µM (experimental groups). Primary osteoblasts were incubated with 5-HT for 2 days, and the proliferation rate was measured. |

Applications |
Growth of osteoblasts treated with serotonin (1 nM-10 µM) was inhibited compared to controls. This influence was alleviated in the 10 µM and 1 µM groups (relatively high concentrations). |

Preparation method |
Twenty-five adult female C57BL/6 mice were divided into five groups. Colon tumours in mice were induced by AOM and DSS. Respectively, 10 mg/kg 5-HT was intraperitoneally injected (5-HT group), 20 mg/kg citalopram in normal saline was administered intragastrically (citalopram group), and then both 5-HT and citalopram were administered in combination (combined group). Following the procedure, the mice were sacrificed after 90 days, and their blood serum was collected to quantify 5-HT contents. The tumours were separated from each animal, and their sizes and volumes were measured. |

After 90 days of treatment with 10 mg/kg 5-HT, the tumour number and volume in the mice’s intestinal mucosa were 11.60 ± 1.52 and 14.35 ± 2.72 mm3, respectively. 5-HT promotes primary colon carcinogenesis in mice. |

5-hydroxytryptamine, (5-HT, Serotonin) a neurotransmitter, is a key messenger that mediates several central and peripheral functions in the human body[1]. 5-HT is also a catechol O-methyltransferase (COMT) inhibitor with a Ki of 44 µM[2].
In vitro, 5-Hydroxytryptamine (1 nmol/L-10 micromol/L) caused vasoconstriction in a concentration-dependent manner[3]. In vitro, 10 μM 5-HT proliferates colon cancer cells through enhancing YAP (Yes-associated protein) expression[1]. In vitro, for left lower lobe of lungs isolated from the dog, infusion of 5-hydroxytryptamine (100 micrograms/min) increased the pulmonary pre- and post-capillary resistances, but did not change the filtration coefficient[4].
In vivo, topical 5-HT (1-10 nmols) or the 5-HT1/7 receptor agonist, 5-carboxamidotryptamine (10-30 nM), dilated third- and fourth-order arterioles, responses that were abolished by 1 μM SB269970, a selective 5-HT7 receptor antagonist[5].

在体内,局部5-HT(1-10 nmols)或5-HT1/7受体激动剂5-甲酰胺色氨酸(10-30 nM)扩张第三和第四级小动脉,反应被1μM SB269970(一种选择性5-HT7受体拮抗剂)消除[4]。
