A high-performance liquid chromatography with circular dichroism detector for determination of stereochemistry of 6, 9-oxygen bridge dibenzocyclooctadiene lignans from kadsura coccinea

The stereochemistry of two 6, 9-oxygen bridge dibenzocyclooctadiene lignans from Kadsura coccinea, are difficult to separate and very unstable. The present study was designed to develop a high-performance liquid chromatography using circular dichroism detection for the analysis of the stereochemistry. A new 6, 9-oxygen bridge dibenzocyclooctadiene lignans named Kadsulignan Q was firstly found with an S-biphenyl configuration. The other compound was identified as Kadsulignan L with an R- biphenyl configuration. In order to obtain kinetic data on their reversible interconversion, the stability was measured at different deuterated solvents such as deuterated methanol, deuterated chloroform and deuterated dimethylsulfoxide. The lignans were more unstable and converted more easily in deuterated methanol than in deuterated chloroform and deuterated dimethylsulfoxide.

Lignans from Kadsura angustifolia

A new dibenzocyclooctadiene lignan named angustifolin D (1) together with four known lignans: Kadsulignan L (2), kadsulignan N (3), schisantherin P (4) and meso-dihydroguaiaretic acid (5) were isolated from the stems of Kadsura angustifolia. Their structures and stereochemistries were elucidated by spectral studies. Compounds 2 and 5 showed moderate platelet-activating factor (PAF) antagonistic activities with IC50 values of 2.6 x 10(-5) and 4.1 x 10(-5) M, respectively.

Dibenzocyclooctane lignans from the stems of Kadsura induta and their antiviral effect on hepatitis B virus

Two new dibenzocyclooctane lignans, kadsurindutins A (1) and B (2), were isolated from the stems of Kadsura induta, together with the known, structurally related lignans schisantherin L (3), schisantherin P (4), Kadsulignan L (5), and neokadsuranin (6). Their structures and configurations were elucidated by spectroscopic methods (UV, ORD, CD, IR, 1D- and 2D-NMR) in combination with mass-spectrometric (HR-MS) techniques. Compounds 1, 5, and 6 showed in vitro antiviral effects on hepatitis B virus.

[Studies on chemical constituents of Schisandra propinqua (Wall.) Hook. f. et Thoms]

Objective: To isolate and characterize compounds from the stems of Schisandra propinqua. Method: Extracting with solvent, isolating by column chromatography and identifying by the spectroscopic methods. Result: Six dibenzocyclooctadiene lignans were isolated and identified as tigloylgomisin P(1), angeloylgomisin O(2), angeloylisogomisin O(3), Kadsulignan L(4), (+/-) 5,8-epoxyl-6, 7-dimethyl-2',3',2",3"-dimethylenedioxy-4', 1"-dimethyl-1,2:3,4-dibenzo-1, 3-cyclooctadiene(5) and wuweizisu C(6). Conclusion: Compounds 4 and 5 were the first two dibenzocyclooctadiene lignans with an 6,9-epoxy bridge cycle discovered in the genus Schisandra. The others were originally isolated from S. propinqua.
