Influence of Gardenia yellow on in vitro slow starch digestion and its action mechanism

This study aimed to explore the influence of Gardenia yellow on in vitro wheat starch digestion. The influence of Gardenia yellow on the digestion properties of starch was determined through in vitro digestion, and its action mechanism on slow starch digestion was revealed by laser scanning confocal microscopy, enzymatic inhibition dynamics, and other means of characterization. Results showed that Gardenia yellow could inhibit starch digestion, significantly increase the resistant starch and slowly digestible starch contents in starch (P < 0.05), and trigger the decrease in glycemic and hydrolysis indices. Furthermore, Gardenia yellow could spontaneously bind to the catalytic sites of α-amylase and α-glucosidase, affect their secondary structures through vdW force and hydrophobic interaction, and reduce their catalytic abilities to inhibit the digestion process of wheat starch. Therefore, the interactions of Gardenia yellow with starch and digestive enzymes jointly promote the slow digestion of starch.

Gardenia jasminoides Ellis: Ethnopharmacology, phytochemistry, and pharmacological and industrial applications of an important traditional Chinese medicine

Ethnopharmacological relevance: Gardenia jasminoides Ellis is a popular shrub in the Rubiaceae family. The desiccative ripe fruits of this plant (called Zhizi in China) are well known and frequently used not only as an excellent natural colourant, but also as an important traditional medicine for the treatment of different diseases, such as reducing fire except vexed, clearing away heat evil, and cooling blood and eliminating stasis to activate blood circulation. It has also been declared as the first batch of dual-purpose plants used for food and medical functions in China. Aim of the study: This review aims to provide a critical and systematic summary of the traditional uses, ethnopharmacology, phytochemistry, pharmacology, toxicity and industrial applications of Gardenia jasminoides Ellis and briefly proposes several suggestions for future application prospects. Materials and methods: The related information on Gardenia jasminoides Ellis was obtained from internationally recognized scientific databases through the Internet (PubMed, CNKI, Google Scholar, Baidu Scholar, Web of Science, Medline Plus, ACS, Elsevier and Flora of China) and libraries. Results: Approximately 162 chemical compounds have been isolated and identified from this herb. Among them, iridoid glycosides and yellow pigment are generally considered the main bioactive and characteristic ingredients. Various pharmacological properties, such as a beneficial effect on the nervous, cardiovascular and digestive systems, hepatoprotective activity, antidepressant activity, and anti-inflammatory activity, were also validated in vitro and in vivo. Moreover, geniposide and genipin are the most important iridoid compounds isolated from Gardenia jasminoides Ellis, and genipin is the aglycone of geniposide. As the predominant active ingredient with a distinct pharmacological activity, genipin is also an outstanding biological crosslinking agent. Gardenia yellow pigment has also been widely used as an excellent natural dye-stuff. Hence, Gardenia jasminoides Ellis has been applied to many other fields, including the food industry, textile industry and chemical industry, in addition to its predominant medicinal uses. Conclusions: According to this review, Gardenia jasminoides Ellis is outstanding traditional medical plant used in medicine and food. Pharmacological investigations support the traditional use of this herb and may validate the folk medicinal use of Gardenia jasminoides Ellis to treat different diseases. Iridoid glycosides are potential medicines. Gardenia yellow pigment has been the most important source of a natural colourant for food, cloth and paint for thousands of years. This herb has made great contributions to human survival and development. Moreover, it has also achieved outstanding progress in human life and even in art. Although Gardenia jasminoides Ellis has extremely high and comprehensive utilization values, it is still far from being completely explored. Therefore, the comprehensive development of Gardenia jasminoides Ellis deserves further analysis.

Stability evaluation of Gardenia yellow pigment in presence of different phenolic compounds

Gardenia yellow pigment (GYP) may undergo chemical degradation under different conditions resulting in color fading. This study investigated the effects of different phenolic compounds (caffeic acid, rosmarinic acid, tannic acid, epicatechin, chlorogenic acid, epigallocatechin, and epigallocatechin gallate) on the physical and chemical stability of GYP under light and different temperatures. Furthermore, food models with GYP/phenolic compounds were simulated to evaluate the GYP stability under different cooking methods. The addition of phenolic compounds, especially tannic acid, epigallocatechin gallate, epigallocatechin, and rosmarinic acid, significantly improved the GYP stability during light and thermal treatments. Fourier transform infrared (FTIR) and nuclear magnetic resonance (NMR) spectroscopy confirmed that the formation of hydrogen bonds between GYP and selected phenolic compounds (tannic acid, epigallocatechin gallate, epigallocatechin, and rosmarinic acid), which may lead to the enhancement of GYP stability. Moreover, these selected phenolic compounds provided potent protective effects on GYP under different cooking methods.

Stability evaluation of Gardenia yellow pigment in the presence of different antioxidants or microencapsulating agents

The chemical instability of Gardenia yellow pigment (GYP) limits its utilization in the food industry. In this study, the effects of different antioxidants (0.2% of tea polyphenols, sodium phytate, potassium citrate, and ascorbic acid) and microencapsulating agents (gum Arabic, maltodextrin, inulin, and gum Arabic/maltodextrin) on the degradation of GYP under different conditions (heat, light, and ferric iron) were evaluated. Then, the characteristic properties of microcapsules coated with gum Arabic/maltodextrin, gum Arabic/maltodextrin/tea polyphenols, maltodextrin, and maltodextrin/tea polyphenols were investigated. Furthermore, food models were simulated to evaluate the GYP stability of the microcapsules. The results showed that tea polyphenols, maltodextrin, and gum Arabic/maltodextrin significantly improved the GYP stability. Moreover, the presence of GYP in microcapsules was confirmed by nuclear magnetic resonance and Fourier transform infrared spectroscopy. In addition, GYP-MD/TP possessed high thermal stability under different cooking methods. PRACTICAL APPLICATION: Gardenia yellow pigment (GYP) is easily degraded under light and high-temperature conditions, which limits its applications in the food industry. This study will provide effective clues for expanding the practical applications of GYP in the natural pigment industry.

Preparation of multiresponsive hydrophilic molecularly imprinted microspheres for rapid separation of Gardenia yellow and geniposide from gardenia fruit

In this work, a robust method for the separation of Gardenia yellow and geniposide from gardenia fruit was developed based on a molecularly imprinted solid phase extraction (MISPE) procedure. First, hydrophilic molecularly imprinted microspheres (HMIMs) were prepared using Gardenia yellow as the template via reversible addition fragmentation chain transfer (RAFT) precipitation polymerization. The resultant HMIMs demonstrated the multiresponsiveness to pH, temperature, and magnetism, achieving controllable uptake and release of Gardenia yellow and easy recovery by external magnets. Meanwhile, the HMIMs possessed high adsorption capacity, fast binding kinetics, specific recognition, and reusability. Finally, a MISPE approach using HMIMs as adsorbent was developed for extraction of Gardenia yellow and purification of geniposide after optimization of the adsorption and elution conditions. Thus, efficient separation of Gardenia yellow and geniposide with relative purities of 99.77 ± 0.05% (94.04 ± 0.10% recovered) and 94.50 ± 0.62% (95.40 ± 0.86% recovered), respectively, was achieved.
