Description

Chlorophenol Red β-D-Galactopyranoside is a long-wavelength dye, which can be detected with an absorbance at 570nm[1]. Chlorophenol Red β-D-Galactopyranoside has been widely used to detect β-galactosidase in bacteria and plants [2-3]. The anti-HIV activity of MAGI cells can be clearly detected by Chlorophenol Red β-D-Galactopyranoside with high sensitivity using standard spectrometers[4]. Chlorophenol Red β-D-Galactopyranoside showed high accuracy, allowing detection of enzyme activity at concentrations as low as 0.05mU, independent of blood contamination[5].

实验参考方法

基于Chlorophenol Red β-D-Galactopyranoside的β-半乳糖苷酶检测大肠杆菌方法:

1.Chlorophenol Red β-D-Galactopyranoside溶液配制:

将Chlorophenol Red β-D-Galactopyranoside(0.02至2单位/mL,1单位定义为pH7.3、37℃条件下每分钟水解1μM邻硝基苯-β-D-半乳糖苷生成邻硝基酚和D-半乳糖的酶量)溶于100mM磷酸盐缓冲液(pH6.8),添加50mM巯基乙醇和1mM氯化镁。将4wt%聚-L-精氨酸(pR)溶于双蒸水。配置完成后使用0.22μm滤膜过滤除菌。

2.细菌培养:

所有ATCC大肠杆菌25922菌株从已知细胞密度的甘油冻存管启始培养。制备0,1,10,100,1,000及10000cfu/mL六个浓度梯度的250μL培养样本,在含1mM IPTG和25μM的Chlorophenol Red β-D-Galactopyranoside+1wt% pR的胰蛋白酶大豆肉汤(TSB)中培养16小时。

3.吸光度与荧光测定:

37℃培养过程中同步监测信号变化,确定各初始菌浓所需的最短检测时间。采用酶标仪每10分钟读取一次数据(每次读数后震荡混匀),设置参数如下:荧光检测:激发波长580nm(带宽9nm),发射波长610nm(带宽20nm),增益设为最优值。吸光度检测:570nm波长,同时使用OD600值监控细菌生长情况。
