Description

1,2-Dimyristoyl-sn-glycero-3-PC is a synthetic phospholipid[1]. 1,2-Dimyristoyl-sn-glycero-3-PC is usually used to construct liposomes for the study of lipid monolayer and bilayer structures and drug delivery systems[2][3]. 1,2-Dimyristoyl-sn-glycero-3-PC can also be used in metabolism-related research[4].

In vitro, 1,2-Dimyristoyl-sn-glycero-3-PC (200μg/mL; 24h) significantly enhanced PVP-SPIONs uptake in PC-12 cells[5]. 1,2-Dimyristoyl-sn-glycero-3-PC (0.08-250μM; 72h) increases Carmofur stability and antiproliferative effect in C6 glioma cells[6].

In vivo, 1,2-Dimyristoyl-sn-glycero-3-PC (2-2.5mg/day/mouse in drinking water; 8 weeks) significantly reduced aortic sinus lesion area and en face whole aorta lesion area in 6-month-old male apoE-null mice[7].

Preparation Method

1,2-Dimyristoyl-sn-glycero-3-PC vesicles were prepared as follows: Carmofur (26mg, 0.1mmol) and 1,2-Dimyristoyl-sn-glycero-3-PC (0.334g, 0.5mmol) were dissolved in 25ml of chloroform and evaporated on rotatory evaporator under reduced pressure with slow rotation without any heating. Then additional portion of chloroform was added and the procedure was repeated for 5 times. C6 glioma cells were grown in standard medium Dulbecco’s modified Eagle’s medium (DMEM) supplemented with 10% fetal bovine serum. About 1-2×104cells/well were placed in 96-well plates immediately after 1,2-Dimyristoyl-sn-glycero-3-PC vesicles (250, 50, 10, 2, 0.4 and 0.08μM) were added to the wells. The plates were incubated for 72h, at 37℃, 5% CO2. The number of surviving cells was determined using MTT assay. The control cells were cultured on separate plate and an appropriate volume of control buffer was added to control cells. Each assay was conducted in triplicate.

Animal models

Male and female apoE-null mice

Preparation Method

Male and female apoE-null mice were maintained on a chow diet. The 1,2-Dimyristoyl-sn-glycero-3-PC, obtained in the form of a fine powder, readily resulted in a uniform suspension when mixed in water for 1mg/mL. The suspension was added to 50-mL graduated polypropylene tubes fitted with standard rubber stoppers and stainless steel tubes of a type used routinely in mouse water bottles. 1mg/mL 1,2-Dimyristoyl-sn-glycero-3-PC in the drinking water was given to mice for 8 weeks. The tubes were observed daily, and the lipids were found by inspection to remain in suspension. The contents of the tubes were changed every other day. The mice drank 2 to 2.5mL/d per mouse, and there was no significant difference in the amount of suspension consumed between the groups. Aortic lesions were scored by histopathology analysis.
