Animal experiment:

Mice[2]Nulliparious female mice weighed 25-28 g (8-9 weeks) are used throughout the study. The mice are ddY strain and specific pathohen-free. As ovulation can be induced in mice at any time with administration of human chorionic gonadotrophin (hCG), 5.0 IU of hCG is injected subcutaneously into each mouse at 17:00, irrespective of the stage of the estrus cycle. This day is designated as Day 0. Nine groups are established according to the dose of Fertirelin ranging from 0.001 to 1.0 μg, and the administration are carried out at 22:00 on Day 0 and at 2:00, 6:00, 10:00 and 14:00 on Day 1[2].

Fertirelin, an analog of GnRH 2.5 to 10 times more potent than native GnRH , has become the treatment of choice for reversing follicular cysts[1]. Administration of high doses of Fertirelin is found to reduce superovulatory responses[2].

Influence of GnRH analogue (Fertirelin acetate) doses on synchronization of ovulation and fixed-time artificial insemination in lactating dairy cows

The effect of using a dose of 50 micro g rather than 100 micro g Fertirelin in an ovulation/fixed-time insemination protocol for Holstein-Friesian dairy cows was investigated in three experiments. In each experiment, Fertirelin was administered at the beginning of the protocol followed 7 days later by 500 micro g cloprosterol. Two days later, a second dose of Fertirelin was given and AI performed 16-19 h later regardless of the incidence of behavioral oestrus. The effect on conception rate was studied in experiment 1 using 114 postpartum anoestrus cows. There was no significant difference in the age, parity or number of days after parturition in each treatment groups. The conception rate did not differ between the 50 micro g Fertirelin group (61.1%; n=72) and the 100 micro g group (59.5%; n=42; NS). In experiment 2, a further 12 cows at 40-60 days postpartum were treated with 100 or 50 micro g Fertirelin (n=6 per dose) with treatment commencing in the follicular or luteal phase of the oestrous cycle. The plasma concentration of luteinizing hormone (LH) reached similar peaks of over 5 ng/ml 120 min after the intramuscular administration of Fertirelin in both groups. There were no significant differences in LH levels between treatments or phase of the oestrous cycle when treatment commenced. Doses of 50 and 100 micro g Fertirelin were compared in experiment 3 using 17 cows to study follicular wave development and synchronization by transrectal ultrasonography, conception rate and corpus luteum function. There were no significant differences between treatments for these factors. It was concluded that using a dose of 50 micro g Fertirelin enabled the drug costs to be reduced without affecting the efficiency of a synchronization of ovulation/fixed-time AI protocol for dairy cows.

Fertirelin and buserelin compared by LH release, milk progesterone and subsequent reproductive performance in dairy cows treated for follicular cysts

This field study compared the efficacy of a single injection of 200 microg Fertirelin with that of 20 microg buserelin in shortening the recovery period of 68 laotating Holstein-Friesian cows with ovarian follicular cysts 65 d post partum or later. Differential diagnosis was based not only on palpation per rectum but also on skim milk progesterone profiles (or= 1.0 ng/ml) 1 wk after treatment, was evidenced in 75% of the Fertirelin group and 72% of the buserelin group. A 74% conception rate was achieved in fertirelin-treated cows, with a mean interval of 71 d from treatment to conception. In the buserelin-treated cows, 65% conceived and the treatment-to-conception interval was 63 d. Differences were insignificant. At the doses used the two GnRH analogs were deemed equally effective in managing particularly stubborn follicular cysts.

Effect of Fertirelin acetate or buserelin on conception rate at first or second insemination in lactating dairy cows

The objective of this study was to determine if Fertirelin acetate and buserelin, two GnRH agonists, improve conception rate when administered at the time of first or second AI in lactating dairy cows. The study consisted of a common protocol conducted at 10 commercial dairy farms. Approximately 150 cows within each dairy were assigned randomly in replicates to receive intramuscularly either no injection or injection of 25, 50, 75, or 100 micrograms Fertirelin acetate or 10 micrograms buserelin immediately after AI. Cows were subjected to the reproductive management practices normal for each location. Cows at each location were palpated for pregnancy status at 35 to 60 d postinjection. No improvement in conception rate was detected in response to either agonist (control = 48%, Fertirelin acetate = 41.5%, buserelin = 39.7%). Conception rate was unaffected by either days postpartum at injection or overall fertility of the individual herds. These observations do not support the routine use of doses of 25 to 100 micrograms Fertirelin acetate or 10 micrograms buserelin at the time of first or second AI as a means to improve conception rate in lactating dairy cows.

LH and FSH response of Holstein heifers to Fertirelin acetate, gonadorelin and buserelin

This study was conducted to describe the changes in serum LH and FSH concentrations in Holstein heifers following intramuscular (i.m.) injection of various dosages of Fertirelin acetate and other commercially available GnRH products at their labeled dosages. The design was an incomplete Latin-square which gave nine replicates of each treatment. Treatments administered on Days 8 to 16 of the estrous cycle consisted of saline; 25, 50, 100 or 200 microg Fertirelin acetate; 100, 250 or 500 microg gonadorelin; and 10 or 20 microg buserelin. Blood samples were collected via jugular catheters from 1 h before to 8 h after each injection. Log (Base 2) area under the LH and FSH curves (log AUC) were used to evaluate response to Fertirelin acetate dosages and to determine difference (LSD: 0.05) and bioequivalence (alpha = 0.05) between the various dosages of GnRH products tested. Significant quadratic dose response relationships were detected for the LH and FSH log AUC data. Plots of the LH log AUC but not the FSH log AUC data suggested that a response plateau was being approached at the higher dosages of Fertirelin acetate. Bioequivalence (alpha = 0.05) was based on the assumption that two means are equivalent if they differ by no more than 20% of the reference log AUC mean. Using these criteria Fertirelin acetate is 2.5 to 10 times more potent than gonadorelin, whereas buserelin is approximately 10 to 20 times more potent than Fertirelin acetate in the bovine for LH and FSH release.

Effect of fenprostalene 14 days after Fertirelin treatment on intervals from treatment to conception in cows with follicular cysts diagnosed by milk progesterone test

Of 170 cows with ovarian cysts which were detected by palpation of the ovaries per rectum, 88 cows (51.8%) were diagnosed as having follicular cysts on the basis of milk progesterone concentrations. Eighty-three of the 88 cows were used for treatment trials. A group of 56 cows injected subcutaneously with fenprostalene, prostaglandin F2 alpha analog, 14 days after treatment with Fertirelin, an analog of gonadotropin-releasing hormone, compared with another 27 cows treated only with Fertirelin (controls), showed a higher pregnancy rate within 100 days after the initial treatment (66.1 vs 48.1%) and shorter intervals from the treatment to conception (30 +/- 21 vs 43 +/- 27 days). It was concluded that the administration of fenprostalene following Fertirelin treatment is effective in shortening the interval from treatment to conception in cows with follicular cysts.
